Two Gram-positive, moderately thermophilic, endospore-forming, rod-shaped, motile bacteria, designated PDF25(T) and PDF30, were isolated from Karakoc hot spring in the province of Izmir, Turkey, and were characterized in order to determine their phylogenetic positions. 16S rRNA gene sequence analysis revealed that the two strains belonged to the genus Brevibacillus; strain PDF25(T) showed highest sequence similarity to strain PDF30 (99.40/) and Brevibacillus thermoruber DSM 7064(T) (98.5 %). The major fatty acids of strain PDF25(T) were iso-C-15:0 (39.30%), anteiso-C-15:0 (26.10%) and iso-C-16:0 (14.75%). Polar lipid analysis reveal . . .ed diphosphatidylglycerol, phosphatidylglycerol, phosphatidylmonomethylethanolamine, phosphatidylethanolamine, and a variety of unidentified aminophospholipids, phospholipids and aminolipids. The major isoprenoid quinone was MK-7. The G + C content of the genomic DNA was 56.09 mol%. DNA DNA hybridization experiments revealed 58% relatedness between strain PDF25(T) and B. thermoruber DSM 7064(T). Based on these data, the two strains are considered to represent a novel species of the genus Brevibacillus, for which the name Brevibacillus aydinogluensis sp. nov. is proposed. The type strain is PDF25(T) (=DSM 24395(T)=LMG 26289(T)) Daha fazlası Daha az

A new series of mono Mannich bases of 4'-hydroxychalcones 2a-e carrying a variety of aryl groups was synthesized and the in vitro cytotoxic activities of the new compounds were screened against PC-3 cell lines. Bioactivities of 2a-e, which are reported for the first time in this study, were compared against their precursor 4'-hydroxychalcones la-e. Compound 2b was found to be the most potent (IC50 = 3.7 mu M) among the compounds synthesized. In addition, the compounds la-c and 2d showed moderate cytotoxicity. Incorporation of the 3'-piperidinomethyl group in 1b and id raised the potency by 1.68 and 2.19 times respectively and, there . . .fore, seemed to be a noteworthy molecular modification. Correlations were noted between cytotoxicity and one or more physiochemical constants of the aryl ring as well as log P values for the compounds 2a-e. The significant improvement of cytotoxicity of 2b, 2d, and 2e against PC-3 cell lines compared with their chalcone precursors suggests that the incorporation of a piperidinomethyl group is a useful molecular modification and further development of these compounds as candidate cytotoxic agents may be warranted Daha fazlası Daha az

Saccharomyces cerevisiae, industrial yeast isolate, has been of great interest in recent years for fuel ethanol production. The ethanol yield and productivity depend on many inhibitory factors during the fermentation process such as temperature, ethanol, compounds released as the result of pretreatment procedures, and osmotic stress. An ideal strain should be able to grow under different stress conditions occurred at different fermentation steps. Development of tolerant yeast strains can be achieved by reprogramming pathways supporting the ethanol metabolism by regulating the energy balance and detoxicification processes. Complex ge . . .ne interactions should be solved for an in-depth comprehension of the yeast stress tolerance mechanism. Genetic engineering as a powerful biotechnological tool is required to design new strategies for increasing the ethanol fermentation performance. Upregulation of stress tolerance genes by recombinant DNA technology can be a useful approach to overcome inhibitory situations. This review presents the application of several genetic engineering strategies to increase ethanol yield under different stress conditions including inhibitor tolerance, ethanol tolerance, thermotolerance, and osmotolerance Daha fazlası Daha az

The objective of this study was to evaluate the effects of twelve plant growth-promoting rhizobacteria (PGPR; Bacillus mycoides FD07, B. sphaericus RC12, B. pumilus RC19, B. cereus RC18, Variovorax paradoxus RC21, Paenibacillus polymyxa RC35, Pseudomonas putida RC06, B. megaterium RC07, B. megaterium M-3, B. licheniformis RC08, B. subtilis RC11, and B. subtilis OSU-142) used as biofertilisers, on various enzyme activities [glucose-6-phosphate dehydrogenase (G6PD); 6-phosphogluconate dehydrogenase (6PGD); glutathione reductase (GR); and glutathione S-trarisf6r#se (GST)] and on seedling growth in spinach (Spinacia oleracea L.). Enhanc . . .ed plant growth could result from rhizobacterial production of indole-3-acetic acid (IAA). The highest IAA-producing rhizobacteria (RC35 and RC06) produced the highest root and shoot weights. PGPR improved N and P nutrition in spinach, and therefore stimulated plant growth and key enzyme activities. The responses to inoculation, compared to uninoculated control plants, were: -1.9% to +36.4% for shoot fresh weights (FWs), -5.5% to +30.1% for root FWs, -3.5% to +29.8% for shoot dry weights (DWs), -3.8% to +38.5% for root DWs, and -5.9% to +30.1% for leaf areas. Plant growth responses were variable and dependent on the inoculant strain used, as well as on the enzyme activity and growth parameter being evaluated. Close correlations between plant shoot growth, PGPR inoculation, and G6PD (r = 0.28*), 6PGD (r = 0.55*), GR (r = 0.73), and GST (r = 0.64) enzyme activities in spinach have been demonstrated Daha fazlası Daha az

Regenerative Medicine and Tissue Engineering has been gaining critical importance in the medical sector, specifically in cancer and organ-tissue losses. Laboratory studies have been conducted on tissue and organ generation using whole organ decellularization and recellularization techniques. In this study, whole rat pancreas has been decellularized by perfusion and imaged using confocal microscopy. Whole rat pancreas tissue has been divided into control and exerimental groups and decellularized using hypotonic, hypertonic and detergent solutions. The tissues, imbedded in Tissue Tech and frozen at -20 , have been prepared as 30 micro . . .meter sections, dyed using DAPI for confocal microscopic imaging of the cell nuclei. It has been demonstrated that the cell nuclei imaged in the control group have been removed as a result of the decellularization process. These findings indicate that the collagenous tissue has been preserved to a large extent. With the imaging of the serial sections, 3D reconstruction of decellularized pancreas can be obtained. © 2017 IEEE Daha fazlası Daha az
